I agree with Suzanne Gebhart with the addition that the mixture of water diluted Ficoll mixed wit Metrizoate (an X-ray contrast material of high density) should be checked for specific gravity. For PBMC separation this should be 1.077 g/ml (see for details Boyum's article 1968)

Alternatively ready to use Lymphoprep solution is recommended but this is more expensive.
