However, If I were you I would do nested Conventional PCR and then go to the QPCR. But can you please tell me if the 1ng is the total extract (nanodrop measurement) or specific viral load reading?

1ng is the result from spesific viral load reading, Sir. For the measurement, I used RNA Dye Working

Yes you used Qubit Most likely. I think that your concentration is very low. but try to do QPCR for once. you can try viral culture to increase your template load.
