Hello, I would like to ask if there is any assay or technique (any protocol - maybe f.e. flow cytometry or by using a UV vis/ fluorescent spectrophotometer) that measures the quantity of a particular drug (in my case Zinc-Phthalocyanine) within the cells (cancer cell line, adherent) without using Mass spectrometry? I would like to quantify the amount of drug inside of receptor-stimulated cells.
If the drug showed specific absorbance (single peak), you can use it for quantitation.
Actually, you can perform a quantification by absortion and/or emission spectra in sobrenadant combine with flow cytometry assay for cellular content (most quantification by flow cytometry would be semi with arbitrary units unleas you perform some calibration procedure previosly to properly quantify). Zinc Phthalocyanine has identifiable absorption and emission spectra. You can perform a semi-quantification comparing unstimulated and stimulated cells.
