Dear all,

I am looking to generate a construct whereby my tag of interest (GFP) is located at the ER. I would like it to mimic an ER resident protein that has a type 1 orientation (though not in correct terms) where essentially the GFP is 'tethered' to the ER but facing the cytoplasmic side. From what I have gathered is that  generating a: Signal-sequence-GFP-KDEL will locate it to the ER but within the lumen. 

Does anyone have any experience or knowledge of whether this approach of GFP facing the cytosol attached to the ER is possible?

Will the signal sequence determine this?
