ELISA with buffers to mimic the intracellular environment?

I would like to measure binding of proteins that physiologically occurs intracellularly. Is it possible to make a buffer that mimics the cytosolic environment and use that in an ELISA?

I have come across this:

110mM Na Gluconate

25mM KCl

5mM NaCl

2mM MgCl2

2mM Mg EGTA

10mM HEPES to pH 7.14

2.7 mM DTT

I suggest you reconsider the use of a reducing agent (DTT), since immunoglobulins are held together by disulfide bonds.
