It is necessary to have a plate shaker platform when you perform ELISA because the shaker helps in proper mixing of the molecules by bringing about productive collisions between molecules within the wells of the 96-well plate. This will help in completion of the reaction in each incubation step.

Also, during the washing step it is essential that the non-bound molecules (antibody or antigen) be removed in order to avoid background signal, and this will be done in a proper manner by using the plate shaker platform.

On addition of substrate it is very much required that the 96-well plate be shaken substantially so that the product gets distributed homogeneously throughout the wells in the plate just before taking the absorbance reading.

Therefore, the plate shaker platform forms an important component in the ELISA technique to enable one to obtain accurate readings.

Shaker is necessary while performing ELISA experiments, but if you do not have Shaker in your lab then i'll suggest you to perform with your hand, & if you have gyromixture in your lab you can also use that, by covering the top of 96 well plate & place it on gyromixture & on it for few seconds.... But in both methods please take care of mixing of one well variable into another, do do not shake too much.

For ELISA, plates shaker platform is necessary for proper mixing and reaction. You can use careful genital shaking by the hand until you have the plate shaker platform.
