I recently measured by MTS assay the effects of palbociclib on several breast cancer cell lines but the results appeared to be different from published data. As indicated in the attached graph, my tests used palbociclib from two suppliers, selleckchem and Apexbio, which gave similarly strange curves. These curves suggested only the top dose (20uM) of the drug can block cell growth but not other doses (even 7uM), which contradicts to others' work.

This would not be the first time that previous experiments are not reproducible by other researchers. As a matter of fact, it happens quite frequentely. This raises serious doubts about much of the scientific work that is going on worldwide. In the specific case of palbociclib, there are very powerful forces moving behind its research. These forces are interested in business and not in academic issues. So that it is not surprising that you could not get the "good" results that interested parts publish.
I strongly encourage you to double check and confirm your results. May be, palbociclib is not as good as they want us to believe.

although there are many factors which may cause some differences with the previous reports. To compare your results with others you have to use the same chemical from the same suppliers and follow the experimental procedures as published. One of the critical issues is the calculation and the serial dilutions you have prepared of palbociclib and if you have used the drug as free base or HCl salt. Also you know that the sensitivity of the cancerous cell lines toward a drug or chemical may be affected by time

How many time pass between the drug treatment and MTS assay? Does it match the procedure from other researchers?

I think more likely some optimization needs to be done. We have obtained similar results when incubating with drug for 1-2 days. I would suggest seeding fewer cells per well and extending the drug treatment for a week.
