If I wrapped the 12 well plates after seeding the cells with alaminium foil and put it in the incubator, would that affect the growth of the cells? or would that affect the O2 levels that reach to the cells
Aluminium foil possess a low Oxygen permeability - so I think, that if you wrap your 12 well plate with a lot of aluminium foil your cells will suffer from a lower oxygen level. However, it will never be a Hypoxia status! If there are some chemical effects (like oxidation/reduction) I don't know. I can't imagine that there will be a huge impact on your cells - but of course it depends on your cells!!! I am working with resitant breast cance cell lines.

I have maintained AL-wrapped 12 well plates in incubator for 2 days, If you do not completely cover the entire plate, and just put foil on the lid, it will allow air pass through the lid, and it should be fine.
I am working on A549 cells and transfecting these cells with flourecently labeled siRNA and since it's photosensetive then I have to wrap it with foil for 72 hours.
I would not cover my plate completely with aluminium foil because that is definitely going to stress my cells by disturbing the gaseous exchange. As suggested by Samila just cover the top of the lid not the sides from where gaseous exchange occurs.
