I'm currently running an experiment which involves a treatment with a compound dissolved in DMSO. In other words, After seeding the cells in 96 well plate I will be adding the treatment (already dissolved in DMSO) & then I will add the MTT solution.
What would be the effect of adding this treatment on the reading of MTT?
After treatment on the cells, when you are going to do MTT assay any way, medium will be removed and washed with PBS twice or thrice. I dont think that will affect your reaction.
You should have a control consisting of DMSO alone at the same concentration.
You should have a negative control with cells treated with same concentration and amount of DMSO used for dissolving the compound. After normalizing with negative control, you will get the cytotoxic effect only to the compound
