I want to set up an endotoxemia in vivo model and I plan to use i.p. injections of LPS from E. coli to induce the disease. I ordered the O127:B8 E. coli serotype LPS, but I found an unopened vial of O26:B6 E.coli serotype LPS. Now, the temptation is quite big; shall I start my experiments with this LPS or does E. coli serotype alter things too much, regarding cytokine levels in the plasma and onset of disease? Can anyone with relevant experience help?
The E. coli serotype should not alter things very much as lipid A is the critical component in causing endotoxemia. However, there could be subtle changes associated with differences in the O side chains of the two serogroups. My advice is to stick with a single serotype for your study - either one is acceptable.
My first response would be yes, the serotype could make a difference in the outcome of the model. For in vivo mouse models I use the O11:B4 serotype via IP injection. This serotype has been used for models of endotoxin-induced lung injury and works quite well for inducing cytokines associated with inflammation (see Rojas et al. 2005 Am J Physiol Lung Cell Mol Physiol 28: L333-L341). With whatever serotype you decide to use, I would recommend performing a small dose response study to ensure that your model is working and is delivering the required endpoint(s) that you want to study.
Thank you both for these replies! Regarding setting up the model, I was thinking of a titration of 1-10mg/kg LPS and a 2-8hours time frame. My endpoints will be cytokine plasma levels (TNF-a, IL1b, IL6, IL10), lung inflammation and (possibly) peritoneal cavity inflammation.

Please keep in mind that besides A lipid there are the rfb wxyz that may alter the results.I would, very humble, suggest the author to choose just one strain of the ATCC with all data and realize their studies so that their results might be compared with further author and display results that may be compared with everyone now and in the future.The music has not not too much to do with these comments, I guess, but it is a relief sound that may reinforce our suggestions
