Dear fellow researchers,

I have been trying to stain E-cadherin and N-Cadherin in the breast cancer cell in MDA-MB-231 for quite some time. Ideally, they should be expressed at the cell-cell junctions when the cells grow as clusters as seen in MCF-7 or HBL-100. However, MDA-MB-231 cells do not grow as clusters. They grow as individual cells. After optimization of the protocol, I did stain E-Cad and N-Cad but not at the cell-cell junctons. They were stained each time in the cytoplasm. What i need an opinion on is that, can be just be non-specific binding or is it safe to say that the E-Cad and N-Cad are correctly stained? I have read dozens of papers to get an idea about this but haven't reached any concrete conclusion until now. Kindly refer to the attached images and sugeest if this is correct or not.
