I am working on osteosarcoma cells (U2OS) and am looking for ways to block TGF-Beta signaling pathway (towards Oct-4). Has anyone tried anything related to this experiment? Any recommended product that may work with a great probability? Any suggestions for an experimental design?

Hi Bayu Winata It seems SD-208 is a potent inhibitor of TGF-B R1. Thanks for the help!
Inhibiting the whole branch of SMAD2/SMAD3 pathway, I would use SB43152 (usually at concentrations between 1-10uM depending on the cell type)

I am wondering if the TGFB signaling target that I want to study is happening through non-canonical pathway, then will this work or not.
SB targets the kinase domain of the receptor, so that means it might not work on the non-canonical pathway.
You mention OCT4 as target, (I assume POU5F1?). In hESCs at least, the enhancer is bound by SMAD2/4 complexes.
Do you add the TGFb to your medium or is it autocrine signaling? If you add it, you can ommit the ligand in the medium (and remove the serum if added)
