I am working on powdery mildew on Barley, I prepare life culture and I do the inoculation but after 5 days from inoculation the infection state appear but most the 1st leaf of seedling plant were toasted !. I want a standard protocol for inoculation and infection for powdery mildew on Barley or wheat.

Powdery mildew of Barley was my very interested project when I was working in agricultural researches Directorate.. The best inoculation method is Dusting the conidiospores over floated barley leave pieces on 40 ppm of benzimedazole inside petridishes... You can leave the closed dishes in the growth room (20C &14 hr photoperiod).. mildew pustule start to appear at day 4th or 5th , the latent period (Periof from Inoculation to expression of 50% of the pustules must be considered.. this test is really gave you a good idea about the seedling stage response to Blumeria hordei.
By the way.. An excellent source of HR was selected by this method... The new source of resistance carry Mla13 which I named a super mildew resistant gene... I published many manuscript (3 in Arabic and One in English
