I'm using petunia for protoplast extraction and gene transformation.

It's been subcultured many many times and I guess the plants have been used for more than

3-4 years.

We find it difficult to have the colonies form calli these days.

One says maybe it's because the plants are old.

But our petunia seems to produce new buds well with each subculture.

Does the total age of the plants really affect the vigor and viability to regenerate?
