I have a mix of cd8+ and cd4+ tumor infiltrating t-cells isolated from patient tumor. I am expanding them in vitro in media supplemented with IL-2 at a concentration 5U/ml. I believe that IL-2 supplementation helps in the survival and expansion of T-cells and not activation to cytotoxic Tcells. When i added them to a HLA matched 3D cancer culture, with a 3xhigher IL2 dose (as i did not want to change medium) i found that the cancer cells died. Could it be that the Tcells were activated due to higher IL-2? Or could it be because the Tcell were already active as these are tumor infiltrating lyphocytes that were already primed with the tumor antigen?
