I am doing a research on the protective potential of some synthesized peptides from Hydroxyl radical-induced DNA damage (Gel electrophoresis). And the following is the experiment procedures:
To assess the protective effects of the hydrolysate against DNA damage caused by hydroxyl radicals, the reaction was induced by placing the following reagents in an Eppendorf tube: 5μL of genomic DNA (RAW 264.7 cellline), 2 mM FeSO4, and various concentrations of thepurified peptide from black eelpout hydrolysate. Themixture was then incubated at 37 °C for 30 min, followedby the addition of 4μLof10mMH2O2(Dávalos et al.2004). Finally, the mixture was subjected to 1.0% agarosegel electrophoresis and DNA bands were stained withethidium bromide.
