I have performed 3-5 IF experiments using PP2 in varying concentrations from 3-10 uM and it seems at the higher end of the concentrations I used (5uM and 10uM) half of the cells on my cover slips have a decrease in cell area. I am using SUM159 breast cancer cells and am having a difficult time sifting through the literature available to get an answer. I have seen some results demonstrating that PP2 can inhibit cell spreading but if anybody has any experience in this area your input would be greatly appreciated.
Please do the expt. and measure the cell size to know about it. Do a literature search to find out if anybody may have done similar studies.
Src Family Kinases can affect cell adhesion, and there is a possibility that PP2 will affect cells spreading on a slide. In this case each cell will cover smaller area on a slide giving you an impression of different cell size. You can measure area that is taken by each cell using IF, however to measure actual cell size I would suggest using other methods. Flow cytometry would be the best and quickest option in my opinion. Putting some PP2 on your cells and measuring size with flow cytometry might be easier than going through the literature.
