I would like to extract my fungus metabolites and resuspend the dry extracts with methanol. After that I would like to conduct a disc diffusion assay test using the fungus methanol extract against the Ganoderma. So my concern is, does methanol inhibit the growth of Ganoderma? I am afraid that methanol that causes the inhibition of Ganoderma instead of the fungus I am using. Or DMSO would be a better choice? Thanks.
I thought of running through HPLC and elute the particular peak to test on Ganoderma. Thats why I use methanol. Thanks for the advice. I will try it. What do you think about using DMSO instead? Do you think it's suitable ? Thanks.
Dear Angel, methanol is very volatile and You can decap your Petri dishes for some while and let it to evaporate under laminar flow, then put the cap on the dishes. You shall consider control disc with only methanol especially when you want to close the caps soon- the way I do not recommend as there may be interactions among the materials. Then use control discs with only methanol and let all plates be opened under clean air flow of laminar flow to let methanol go away. so simple.
