Hi Donovan,

You need to always place the direct repeat at the 5' end of the crRNA sequence. Based on the literature I have reviewed, the direct repeat is consistently positioned at this end to facilitate binding with the Cas13 enzyme.

Furthermore, I am currently using this method and have designed all my crRNAs with this consideration in mind.
