Do we need to incubate suspension cell before treatment with drugs (or any stimulants)? As I know for adherent cell, it is required so because it need time to get attached. Does it make any difference if we treat directly without pre-incubation?
Cells either attached or in suspension are usually pre-incubated with serum restricted (low) or serum free isosmotic buffered solutions to establish the cultures into a synchronized phase with less or little cytokines, that could possibly interfere with the drug action or specifically if the target is cytokines or other immunological molecules.
If you are sure that the drug or treatment conditions you are planning to employ do not interfere with the growth media containing serum present in the suspension culture media you can certainly go for direct drug treatment.
And It also depends on what protein or what signal you would like to investigate with your cells,and depend on what kind of cell you are handling with. Seeding is a stressful process for cells which can cause some effect on cell signals, synthesis and production of some cytokines or mediators. Pre-incubating the seeded cells at least 1 hr before treating is better than treating the cells immediately after seeding (in my experience).
