I'm trying to find the IC50 concentration of 5-FU, oxaliplatin, and cisplatin in HT29 cells. While doing the MTT assay, the drug-treated cells (low conc. ones) showed more absorbance than the controls. Is this because the drugs are interacting with MTT? I remove drug containing media before adding MTT. Should I do a PBS wash before adding MTT to remove traces of the drug?
