Does anyone use a plasmid vector suitable for mammalian expression by non-viral transfections, and having the following features:
-GFP following an IRES site and not as a fusion with the gene of interest (insert)
-Puromycin resistance gene
-Biotin tagging system
-suitable for genes about 3kb

The only one somewhat similar I found on Addgene. Puromycin, IRES, RFP, mammalian expression.

Thanks for your search. I will not be using a cre-lox system, which is what that plasmid might be suitable for.
I have looked up on Addgene and not found any suitable ones. So I thought if anyone in this forum personally uses such a plasmid (not listed on Addgene), I could benefit.
Oh, sorry. I have only worked with the general pEGFP with direct fusion to the GFP.
Good luck. Maybe you could insert the IRES and the other desirable regions if you have the casettes.
Hi da.. Yes I use a TRIPZ vector which is suitable for by non-viral transfections. I used Lipofectamine 2000 and it seems to work.. But I have RFP instead of GFP and I don't think that will matter much.. And it did work in HeLa cells..
