After drug treatment, I'm tracking the triglycerides and FFA release in cell culture (adipocytes). However, I'm facing some issues.

I already detected the lipids in the cells but couldn't detect them in the media.

The cells were incubated in regular media (DMEM+10%FBS+P/S). For the protocol, I washed the cells with PBS (2X) and set them with DMEM (1g/L of glucose) + 1% BSA FFA-free for 6h. After this period, I collected the media (basal measurement), washed the cells, and added the drug for 16h. Finally, I collected the media and harvested the cells with NP40 (for TG and FFA measurements). Unfortunately, I did not detect the lipids in the cell culture media, only in the cells (even incubating the cells with Glucagon 20 nM).

Does anyone already perform that protocol and could recommend some tricks?
