I am testing a therapeutic treatment. Since there is a chance that the treatment will not work, I need positive controls for my assays. I am using PrestoBlue to measure redox changes which could reflect changes in cell number or metabolism. I plan to seed wells with different cell numbers and measure differences in fluorescence, but I would also like to show that my assay is sensitive to changes in metabolic rate without a change in cell number. Is there something I can add to the cultures to produce this type of measurable change? I am using fibroblasts derived from tenocytes.
