Studying cell migration of glioblastoma primary cell lines and we're looking for alternatives to FBS to promote adhesion without altering the migration phenotype. Does anyone know the particular components of FBS that promote adhesion?
Hi Mariah, I know lots of things about cell adhesion in FBS.
To begin with, the major protein that sticks to tissue culture plastic is vitronectin (VN). VN on the plastic exposes its RGD sequence, that serves an attachment sequence for integrins avb3, avb5, avb1, etc.
In short, if there is no vitronectin on the TC plastic, there will be no cell adhesion,
You can do a quick experiment to convince yourself. Take a bacterial plate that is non tc treated, add cells and see that they will not adhere and die..
