Please i still want more clarity on synthesis of silver nanoparticles. I tried different parameters like changing concentration of plant extract, for silver nitrate and so on. I observed color change from green to dark brown but yet there is nor SPR at 420. My peak is now at 345. Is it possible that my plant extract cannot reduce silver nitrate or what is the justification? Or maybe the problem could be silver nitrate?
The Ag NP nucleation may be negatively affected by concurrent processes that may depend on the plant extract itself. Even if Ag+ reduction is activated, as it is highly probable with plant extracts, higher velocity processes may inhibit any following Ag0-Ag0 clustering... for example chelating processes involving prphyrin rings surely present in your extract (chlorophyll). Finding the right concentration ranges could be not easy.
However, you can try adding a further experimental parameter, that is UV irradiation (with a common UV lamp) being careful in setting known fixed values of the radiance by first measuring a calibration curve for the lamp (irradiated power vs distance from the lamp). With UV irradiation you will drive the relative velocities of concurrent chemical processes in the solution. That could require many trials (a more or less large matrix of concentration/Uv radiance/time setups), but I think it is interesting and may provide more affordable receipes to produce NPs with specific extracts.
