I have tried several mutations myself as well as for my labmates. My understanding indicates, there is not much difference in two categories of primers as anyway you are going to do selection and sequencing studies later.
Start with anyone category, select multiple colonies, purify and get your DNA sequenced. I am sure you will find few sequences of your desired mutation if you select 3-5 colonies during selection.
Thanks Murli, but our only option is screening so library quality is not essential but highly desirable given the screening effort, so any reduction of bias based on primer purity might me advantageous, provided the primer costs justify the screening costs...
To the best of my knowledge, there is not huge differences in primer quality from these two methods. Considering cost of primer may be a good criteria as irrespective of what primer you use, post-transfection protocol and cost will remain almost same.
Thanks, yes, they compared DNA polymerase suppliers, but not oligos. I was looking for a paper where oligo from company A is compared to that from company B, and where the purity is also assessed, etc...
