I would like to identify the phosphorylation residues of a protein by MS.
I expressed my protein in E. coli and the kinase in mammalian cells. They are good quality proteins and the kinase is active. However, the level of phosphorylation is low. There isn't phospho-specific antibody for this protein, so the kinase assay (30, 60, 120, 240 min) was done using 32P-gATP and the signal was very low even after a week of exposure, compared to other substrates. I am worried that for MS the phosphorylation is not going to be enough for peptide identification.
Someone knows a way how to increase or enrich protein phosphorylation?
Could increase kinase concentration help?
I though on doing many reactions and then purified the phospho-protein by the QIAGEN Kit, but I have not experience with it.
