Hi! I think that the best way will be to transfect your cells with the active form of the Notch receptor. the only issue is that you need to find out what is the isoform that fits best in your model (i.e. in some cells the iper-activation of Notch1 may induce apoptosis).

Another (and cheaper) way is to use EDTA, but it's not specific for the Notch pathway and it's difficult to find the right conditions so I will not suggest it.
