I would like to extract bound phenolic in pigmented rice bran under alkaline and ultrasound condition. I followed the protocol using 2M NaOH for hydrolysis under ultrasound treatment and reducing the pH to 2 by HCl and extracted 3-times with ethyl acetate, somehow, I don't seem to get anything out of my sample. The extract looks clear. Do we have to centrifuge after the addition of ethyl acetate?
I've had experience in bound phenolic extraction from pineapple peel. But, in the method I used, the sample for bound phenolic extraction should be the residue after your sample had undergone solvent/ultrasound condition. Your bound phenolic extraction from pigmented rice bran should be the residue after you have performed solvent extraction beforehand. We could also perform bound phenolic extraction either by acid or alkaline hydrolysis whichever is most suitable for your sample. Hope this helps. Good luck!
