Hello everybody! I've started a new project using SH-SY5Y cells but I have had some problems with cell cuture…they appear small and round, I can’t see neurites and they grow as a mixture of floating and adherent cells. Do you have any kind of suggestions?

These cells sometimes take a while to grow up from cryogenic stock. It can sometimes take up to 2-3 weeks for these cells to appear normal. Because SY5Y are undifferentiated neurons (unless put through differentiating procedures), they will only develop small processes. Here is a picture of what my undifferentiated SY5Y typically look like in culture.

You can check the passage number of your SH-SY5Y. They usually lose their characteristics above passage 20. If your stock is newly purchased, give it time to grow for several days and change the medium (half of the medium) every 2-3 days. In my experience, they grow better and faster with several passages.
