I am doing a degradation experiment using a white rot fungi grown in PDB with yeast extract for 7 days. After which, it's washed with phosphate buffer 4.5. I just want to check the capability of my organism to degrade my target compound in the presence of only an energy source which is glucose. The original growth medium contains 20 g/L of glucose. Now, I am preparing phosphate buffer with 20 g/L of glucose too (basing it from the concentration of the growth media), is this proper or not? what is the ideal glucose concentration in degradation experiments? In fact, glucose concentration is a factor to optimize, the ideal glucose concentration depend of your target compound (quality and quantity) and the used fungus (species and biomass concentration)

glucose concentration in a degradation medium should be added to initiate growth an to boost degradation. Therfore, its concentration depends on the target degradable compound. for keratinous substrates we found 1% glucose is good.
Since the medium is for degradation by white rot fungi, you need to optimize the glucose concentration in the medium. This concentration must be noted and you need to have a control so that you can deduct this concentration after the degradation is completed, because degradation itself can probably produce some monomeric sugars. Many authors have found that standard concentration is 20 g/L glucose, unless you have indications that the specific fungus you use behaves strangely.
