I have a few proteins that I have not been able to purify from BL21(DE3), other E. coli host strains, or their original host P. aeruginosa, due to insolubility. I get great induction of the proteins from T7-based vectors but they are always insoluble. Changing the conditions and tags, as well as fusing the proteins to a solubilizing partner, have not helped. I was wondering whether anyone here has any experience using the "SoluBL21" strain from Genlantis or AMSbio. They claim that this is a strain created by directed evolution in order to produce soluble or toxic protein products. If you have used them, could you please share your experience and let me know if you think it may be worth purchasing these cells for my purposes.
