To understand an effect of protein kinase on substrate transport, an inhibition in transport was seen. Analyzing kinetics, the kinase caused decrease in Km of transporter. Indicating that the kinase actually increases affinity of the transporter for the substrate. Under this condition how is it possible that the kinase would decrease the transport?

My question was - For a transporter, can a DECREASE in its Km value cause a decrease in its activity?
In other words, under a condition when the affinity of a transporter for its substrate is increased, can there be a decrease in its transport activity?
I believe what the previous poster was suggesting was that the substrate would bind so tightly, that it would actually move through more slowly.
