I am trying to prepare spore suspension from 10^5 to 10^7 and inject 20 microliters to Galleria:
1- Spin 1ml of spore suspension to collect spores
2- Re-suspend spores with 20 microliters
My problem is that when I go to pull up the spores, I cannot I feel spore suspension (is too heavy).
How can I solve this problem?

What is the device you use to pull up / inject the spores? Is it fungal or bacterial spores?
Maybe the large quantity of the spores clogs up your pipette tip? I often have this problem, when pipetting spore suspensions. I solve this issue by simply cutting away the very end of the pipette tip. Hope this helps!
Another issue might be, if you spin down the spores to much, so that later, when you try to resuspend the spores in 20 ul the pellet doesn't resuspend.
