I'm wondering if anyone has experience with the FlpIn System by Invitrogen?

I was using lipofectamine to generate stable HEK cell lines of 5 MRP1 mutants, however after screening several hundred single cell colonies (by western blot) there weren't any positive clones. Our lab then bought the HEK FlpIn cells from Invitrogen, and I was able to generate the Wild-Type MRP1 stable HEK cell line within roughly a month by lipofectamine transfection (4ug total of pOG44 and pcDNA5/FRT, at a 9:1 ratio), picking single colonies in 0.2 mg/ul Hygromycin B, expanding, and verifying by Western Blot. I found that there were still several colonies that did not express the protein but were resistant to hygromycin, as well as several positively expressing colonies that were not sensitive to zeocin!

I found a good wild-type clone (I think), however am a little dubious of the system now. I have tried 3 different transfections with my 5 mutants and got some positive colonies (about 20% of those that were resistant to hygromycin were positively expressing my protein), however before I could send them for verification of homogeneity, the clones stopped expressing before I could freeze them! I have been trying to generate these stable cell lines for 9 months now and am feeling exhausted!!!
