As a primary anti-body, I was using anti-myc (rabbit one / sigma) as first anti-body then secondary, gold labeled from BBI (anti-rabbit size of gold depending of what you want to see).

Thanks Anne. I used mouse mAb as first antibody for IEM which was not able to show the localization of target protein. I think because the abundance of the target protein is low (IFA), polyclonal antibody may work.

Polyclonal may work but can aslo be less specific. using a first antibody not too big and then a second labeled with small gold can help.
