When i tested flow antibody titration, I used the same cell, same voltage, same ab but at series of concentration, the percentage of positive cells does not change at some range of conc., but 1) i can still distinguish two distinct populations, 2) the positive population shift to left with lower conc. of antibody.
Dose anyone have the similar experience?

For several antigens at lymphocytes, the mean fluorescence intensity (MFI) gets lower when adding a lower amount of antibody for flowcytometry. This is not a big problem, as long as the populations you want to assess remain clearly distinguishable, proportions of parent population do not change, and you do not want to do quantitative analyses of MFI.
