I've used EdU in a trial of several proliferation assays including BrdU, CellTrace, and a few others. We cultured cells in both RPMI 1640 and DMEM and found that the media made little difference. Our standard RPMI contained 25 mM HEPES, 10% FCS, 1% NEAA, 5x105 mM b2-mercaptoethanol, 100Uml1 penicillin, and 100 ugml1 streptomycin. We settled in 2x105 cells per well in a 96 well format.

For evaluation of lymphocyte proliferation you can use MTT assay. For this :
a)separation of lymphocytes cell from whole blood using ficoll
b) lymphocyte culture in RPMI media
c)Ancobasion for 72h
d) Add 20 microliter of MTT
E)Read OD in 620nm
f)Comparation with control samples.
