Hi all. I'm trying to plastic embed and section embryoid bodies from H9 stem cells, made from 20,000 cells using the immuno-bed kit from polysciences.
These are VERY delicate small tissues. So what is the best step to stop at for storage past 48 hours (ideally up to 4 days) and what methods should be used? The only literature I can find suggests storing near the end of the dehydration step in 70% alcohol. Also, will damage to the tissue be seen if stored at the infiltration step for approximately 4 days?
I want to avoid as much damage as possible, so would it just be ideal to store for no more than 24 hours at each step before plastic embedding?
Thanks in advance for any insight! I only have some slight experience with cryosectioning.
