I work at Center for Advanced research in Plant Tissue Culture, Anand Agricultural University, Anand, India. Currently I am woking on micropropagation of Sandalwood. I have successfully completed multiplication but finding difficulties in rooting phase. I have experimented various approaches for in vitro rooting such as higher auxin levels, combinations of auxins, pulse treatments, various hosts, biotic as well as abiotic stress but none of them have proved viable. The frequency of getting roots is very low (only 1 at every 100). Please suggest a reliable method for high frequency rooting.

also, as against the previous beliefs it is been reported that CK has an equally imprtant role in rooting induction as auxin, please refer to one article "releasing cytokinin brakes on rooting" which featured in journal of plant physiology, this issue or the previous one

Microshoots treated with 20 or 50 mg/l IBA for 48 h produced roots on growth-regulator-free, 1/4 or 1/2MS basal salts medium with vitamin B5 and 2% sucrose followed by one-week incubation in the dark. Also, shoots could produce root after being transferred in WPM medium for 12 weeks.

For rooting of regenerated plants , reducing sugar content in the medium to 10-20 g/l, and/or add low content of auxin, such as IAA 0.1 mg/l; or add additional NH4NO3 10-20 mM to the rooting-medium may help.

We have reviewed various aspects of sandalwood micropropagation. I suspect that you may have used higher concentrations of BA (> 2.5 mg/l BA or > 0.5 TDZ ) for culture initiation and maitainance of shoot multiplication, which effects quality of shoot and you will get pale green or yellowish shoots. Hence, these shoots not good for rooting some times higher concnetrations of these two PGRs cause vitrifications/hyperhydricity of shoots i.e. also not suitable for in vitro rooting. If you have shoots with green stem and green leaves you can try 2 mg/l IBA and 1/2 MS + 1 g/l Activated charcoal for in vitro rooting or you can try exvitro rooting with pulse treatment with 5000 mg/l IBA for 5 mins followed by transfer of shoots to potting misture with soilrite and watered with 1/8 strength MS at regular interval of alternate day (1 day distilled water/2day MS/3rd day distilled water....). Addition of activated charcoal often give good results in promoting roots in woody species... you may try it out!
In vitro rooting at the commercial level requires an optimal protocol, so you should use inexpensive and appropriate treatments. Please, try some of the following ideas on your plant:
1. Use of different auxin hormones (IBA, IAA, NAA and etc) and their combination with very low content of cytokinins
2. The use of low concentrations of abscisic acid promotes rooting in most species
3. Light is an important factor in rooting, so use dark treatment or use blue and red light
4. Use of hormones that are effective in rooting (Jasmonate )
5. Humic acid often has a positive effect on rooting
