I culture isolated mouse islets at 27oC for the first 48h changing the media after 24 and 48h, then change the temp to 37oC (Zmuda 2011 JOVE).  I have cultured islets happily for up to 3-4 weeks in non-TC treated dishes so they dont stick to the culture dish, but I find that they often stick to one another forming large, lobed islets which can develop necrotic cores.  Does anyone have experience with this and perhaps found a way to reduce this issue?
