I am trying to stably express miRNAs in primary murine T cells (from spleen) to adoptively transfere them later into CD4 deficient mice.

Currently, I am working on a protocol to get T cells transduced.We don't want to activate the T cells as it is done in all publications that I could find. I am working with a 3rd gen. lentivirus that expresses the MLV envelope protein for increased safety (we don't need the viruses to be able to infect human cells).

A first trial with spinoculation of total splenocytes using "Viral Entry" from ABM as a transduction enhancer led to poor transduction of CD3- cells and absolutely no transduction of CD3+ cells.

Next, I will use LentiBoost (Sirion) as a transduction enhancer, but I would like to ask first, if anyone has experience with this type of transduction and is willing to share.
