I have isolated some phages from sewage samples and need to determine whether they have RNA or DNA genomes. I understand that I should treat samples with RNAse/DNAse and check for lysis in an agar overlay, or extract the nucleic acid then treat with RNAse/DNAse and run a gel but I am struggling to find a protocol for either of these methods. I also am unsure whether a protease is needed prior to nuclease treatment?
