I want to do NMR for my G-quadruplex forming Oligonucleotides in presence of a specific metal ion. Our lab is new to this topic and no one in our lab has experienced about this.I have tried using some online protocol for NMR sample preparation 2-3 times but it does not work good. Does anyone of you are using this techniques in your lab and if yes, can you please share the protocols for the same.I would be highly obliged.
It is not always possible to record the NMR spectrum in the presence of a particular metal ion.
1- It must have nmr standard of the relevant metal ion
2- The device should be calibrated first with the nmr starter of the metal ion likely to be in the sample.
3- Then the spectrum of the sample should be recorded from the device.

Such analysis by means of NMR is how to say...?! NMR does not appear method of choice.
It is reliable to use mass spectrometry, because of it provides not only qualitative analysis of the coordination capability, but also quantification of different physico-chemical and molecular parameters, as well as, determination of the concentration of the analytes in solution.
Thus, you could pay attention to reference [1] associated with MS of bases, inclusing self-associates. Therein, has been discussed namely, the G-quadruplex. Reference [2] provides you innovative quantitative relations yielding to kinetic, thermodynamic and diffusion parameters of analytes based on mass spectrometry. The reliability of the method shows r =1 at pg.(mL)-1 concentration of the analytes. Such method performances are far beyond the capability of the NMR.
