Hello everyone! I've recently ordered a DuoSet™ ELISA kit from R&D Systems, and while reviewing the protocol, I came across a few points that raised some questions. If anyone has experience with this kit, I'd really appreciate your input.

  • In the general protocol, step 3 instructs to block the plates by adding 300 µL of reagent diluent only. This seems a bit unusual to me—shouldn’t the blocking buffer contain a protein component, such as 2% normal goat serum (cat# DY005), especially since it’s already recommended for the detection antibody dilution? Blocking is typically done with a protein-containing solution to reduce non-specific binding.
  • Also, I noticed that the 5X Reagent Diluent 3 contains 25% Tween. When diluted to 1X, this still results in 5% Tween, which seems quite high. We're more familiar with using lower concentrations of Tween, usually around 0.05–0.1%, primarily for washing steps. Is this high concentration standard for blocking or other steps?
  • Lastly, it seems like the Reagent Diluent 3 provided in the kit isn't very abundant. Has anyone else found this to be limiting?
