I have transformant Saccharomyces, but I'm having trouble to find a good assay for detection of the ACC activity. Does anyone could suggest one?
Have you tested already any method?
I ask because I am no familiar with Saccharomyces but determining ACC activity should not be a problem in, let say, a mammalian cell or mammalian tissue sample.
In any case perhaps this paper (below) may help you AND VERY IMPORTANT, remove from you sample any small molecule, i.e. use just a protein preparation. Otherwise the many samall molecules in the sample will affect the activity determination. Use dialysis or a PD10 desalting column to isolate the high MW molecules and remove the small ones.
