I would like any information on what kind of metal, size, and/or where to buy this. I've been told this will keep the virus at the bottom of the tube when a pellet is not visible. Thanks!
I have done hundreds of lentivirus productions and I have never heard about using anything like that for lentivirus isolation. Can you give me more information about where did you heard about this?
In my experience, if you ultracentrifuge the culture supernatant for 2h at 26000rpm at 4°C, the virus is pelleting in the bottom of the tube for sure. If you don't see a pellet, you are producing very few viruses and should check the transfection steps and the viability of the packaging cells. In any case, you can remove the supernatant and resuspend that "invisible pellet" (which is actually there even when you cannot see it) in cold PBS and titer the solution.
My PI was the one who brought it up so I'm guessing she got the idea from her old lab (she never used it herself). It very well could be that my virus production is low so I'll try modifying those steps. Thanks!
