If anybody knows the analytic procedure for the extraction of free sugar or mono saccharides from sediments or soil ? If anybody have standardized any method to quantify total sugar present ?Any one have any reference paper on this? How to get the sugar analysis procedure by HPLC RI detector?

By above method you can estimate total sugar amount. Than you can try paper chromatography (quality, semiquantity). Also you can try methylation (or acetylation) an analyse it on GC (or GC-MS) (qual. quant.; products of methylation can be extracted). On HPLC there will collide all other material, like salts, acids, ... only if you can purify the sugars..
