Does anybody have used OGA assay ( O-GlcNAcase activity) in tissue sample?

I am looking for O-GlCNAcase enzyme activity in my samples( triple transgenic mice brain tissue) by using Zachara protocol and I avoid the one step of removing lysosomal hexoaminidase and directly desalted my protein to proceed with enzyme assay.I used 25 ug of protein concentration .I didnt find any activity in my samples.So I bit confused that whether the amount of sample is insufficient or the skipped step is responsible for failure of my protocol.Please suggestions are most welcome.
